Description

Simufilam (CAS 1224591-33-6) is a small-molecule modulator of filamin A (FLNA), classified as a spirocyclic lactam compound. Its molecular formula is C₁₅H₂₁N₃O with a molecular weight of 259.35 g/mol. It is also known as PTI-125 and PTI-910.

Simufilam is reported to bind the C-terminal domain of filamin A, restoring its native conformation and attenuating aberrant protein–protein interactions associated with neuroinflammatory and neurodegenerative signaling cascades. This binding is proposed to reduce downstream dysregulation of α7 nicotinic acetylcholine receptors and toll-like receptor 4 complexes. The compound belongs to the 1,4,8-triazaspiro[4.5]decan-2-one chemotype, conferring conformational rigidity relevant to selective target engagement.
