Description

2-(4-Azidophenyl)acetic acid (CAS 62893-37-2) is an aryl azide–functionalized acetic acid derivative used as a click chemistry building block and photoaffinity labeling reagent. Its molecular formula is C₈H₇N₃O₂ with a molecular weight of 177.16 g/mol. It is also known as 4-azidophenylacetic acid and N3-PhAc-OH.

The compound bears a para-azide group on a phenylacetic acid scaffold, enabling participation in copper-catalyzed azide–alkyne cycloaddition (CuAAC) reactions with terminal alkynes to yield stable 1,2,3-triazole linkages. The free carboxylic acid provides a convenient handle for amide bond formation or NHS ester activation, allowing dual-mode conjugation strategies in probe and linker assembly.

  • Bioconjugation via CuAAC for antibody, peptide, and protein labeling
  • Photoaffinity probe construction exploiting aryl azide photochemistry
  • Synthesis of heterobifunctional linkers for PROTAC and ADC assembly
  • Structure–activity relationship studies on azide-bearing phenylacetate scaffolds
