VH298 (CAS 2097381-85-4) is a potent VHL E3 ligase ligand and cell-permeable hypoxia-inducible factor (HIF) stabilizer. Its molecular formula is C₂₇H₃₃N₅O₄S with a molecular weight of 523.60 g/mol. The compound is also known as VH298 in the targeted protein degradation literature and serves as a validated VHL-binding warhead in PROTAC design.
VH298 is reported to bind the von Hippel–Lindau (VHL) protein with high affinity, competitively blocking the interaction between VHL and hydroxyproline-containing HIF-1α peptides. By occupying the VHL substrate-binding site, it stabilizes HIF-α subunits and activates downstream hypoxia-response pathways in normoxic conditions. The compound incorporates a hydroxyproline-containing pyrrolidine scaffold characteristic of optimized VHL ligands.
