Description
Abemaciclib (CAS 1231929-97-7) is a selective, orally bioavailable cyclin-dependent kinase (CDK) inhibitor with molecular formula C₂₇H₃₂F₂N₈ and MW 506.60 g/mol. It is also known as LY2835219 and Verzenio.
Abemaciclib is reported to act as a potent, ATP-competitive inhibitor of CDK4 and CDK6, kinases that phosphorylate the retinoblastoma protein (Rb) to drive G1-to-S phase cell cycle progression. By blocking Rb phosphorylation, it is reported to arrest susceptible cells in G1 and suppress proliferation. Its benzimidazole-pyrimidine scaffold confers selectivity within the CDK family, with notably continuous dosing tolerance relative to other CDK4/6 inhibitors.
