Labuxtinib (CAS 1426449-01-5) is a selective ITK (interleukin-2-inducible T-cell kinase) inhibitor with molecular formula C₂₀H₁₆FN₅O₂ and MW 377.40 g/mol. It is also known as THB335. The compound features an imidazo[1,2-a]pyridine-3-carboxamide core linked to a 1,2,4-oxadiazole bearing a (1R,2S)-2-fluorocyclopropyl substituent.
Labuxtinib is reported to selectively inhibit ITK, a non-receptor tyrosine kinase of the TEC family that plays a central role in T-cell receptor signaling and downstream activation of PLCγ1. By blocking ITK activity, the compound modulates Th2-skewed immune responses and cytokine production, making it a useful tool for dissecting T-cell–mediated inflammatory pathways.
