KSQ-4279 (CAS 2446480-97-1) is a selective USP1 (ubiquitin-specific protease 1) inhibitor with molecular formula C₂₇H₂₅F₃N₈O and molecular weight 534.50 g/mol. It is also known as USP1-IN-1.
USP1 is a deubiquitinase that removes monoubiquitin from PCNA and FANCD2, thereby regulating DNA damage tolerance and the Fanconi anemia pathway. KSQ-4279 is reported to inhibit USP1 with high potency and selectivity, disrupting these deubiquitination events and impairing DNA repair signaling in cancer cell models. Its pyrazolo[3,4-d]pyrimidine core engages the catalytic site of USP1, while the trifluoromethylimidazole and cyclopropylmethoxypyrimidine substituents contribute to binding affinity and selectivity.
