Description
Filgotinib (CAS 1206161-97-8) is a selective JAK1 inhibitor with molecular formula C₂₁H₂₃N₅O₃S and molecular weight 425.50 g/mol. It is also known as GLPG-0634 and GS-6034.
Filgotinib acts as a preferential inhibitor of Janus kinase 1 (JAK1) over JAK2, JAK3, and TYK2, functioning through ATP-competitive binding at the kinase active site. By suppressing JAK1-mediated signal transduction, it attenuates downstream STAT phosphorylation and the transcriptional activation of pro-inflammatory cytokine pathways, including those driven by IL-6, IL-12, and IFN-γ.
