Description
Cobimetinib (CAS 934660-93-2) is a selective, allosteric MEK1/2 inhibitor with molecular formula C₂₁H₂₁F₃IN₃O₂ and MW 531.30 g/mol. It is also known as Cotellic, XL518, and GDC-0973.
Cobimetinib is reported to bind a non-ATP-competitive allosteric site on MEK1 and MEK2, blocking phosphorylation of ERK1/2 and thereby suppressing downstream MAPK pathway signaling. This mechanism confers selectivity within the RAS–RAF–MEK–ERK cascade, making it a well-characterized tool for dissecting oncogenic signaling driven by BRAF mutations or upstream RAS activation.
