Description
BAY1436032 (CAS 1803274-65-8) is a pan-mutant isocitrate dehydrogenase 1 (IDH1) inhibitor with molecular formula C₂₆H₃₀F₃N₃O₃ and MW 489.50 g/mol. It is also known as Bay-1436032.
BAY1436032 is reported to selectively inhibit multiple oncogenic mutant forms of IDH1, including R132H, R132C, R132G, R132L, and R132S, without significant activity against the wild-type enzyme. It acts by binding the mutant IDH1 homodimer interface, suppressing the neomorphic production of the oncometabolite 2-hydroxyglutarate (2-HG) and thereby relieving downstream epigenetic dysregulation associated with IDH1-mutant malignancies.
