AZD 3147 is reported to selectively inhibit both mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2) through ATP-competitive binding at the mTOR kinase domain. By suppressing both complexes simultaneously, it is reported to block downstream signaling through S6K1/4E-BP1 and AKT(Ser473), offering more complete pathway inhibition than rapalog-class allosteric inhibitors. This dual blockade is relevant to PI3K/AKT/mTOR pathway research in oncology and metabolic disease contexts.
