Ravoxertinib (CAS 1453848-26-4) is a selective ERK1/2 inhibitor with molecular formula C₂₁H₁₈ClFN₆O₂ and molecular weight 440.90 g/mol. It is also known as GDC-0994 and RG-7842.
Ravoxertinib is reported to act as a potent, ATP-competitive inhibitor of extracellular signal-regulated kinases 1 and 2 (ERK1/2), the terminal effector kinases of the RAS–RAF–MEK–ERK mitogen-activated protein kinase (MAPK) cascade. By occupying the ATP-binding pocket, it is reported to suppress ERK1/2 catalytic activity and downstream phosphorylation of substrates involved in cell proliferation and survival. Its selectivity profile makes it a useful tool for dissecting ERK-dependent signaling in MAPK-dysregulated cellular contexts.
